Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate SDHB in samples. An antibody specific for SDHB has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySDHB present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SDHB is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SDHB bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Complex II in mitochondria, of which succinate dehydrogenase (EC 1.3.99.1) is a component, has 4 subunits. In order of decreasing molecular mass, they are the flavoprotein (SDHA), the iron sulfur protein (SDHB), and the 2 integral membrane proteins, SDHC and SDHD.
Kita et al. (1990) cloned and sequenced the iron sulfur protein subunit. A clone was isolated from a human liver cDNA library. The open reading frame encodes a 252-amino acid protein. The amino acid sequence showed approximately 94% homology with that of bovine heart. Au et al. (1995) described the complete genomic clone for the gene encoding the iron sulfur protein subunit.
